Quantifying Tissue Cell Surface Antigens by Flow Cytometry

Accelero would like to thank Miltenyi Biotec GmbH for the opportunity to test the gentleMACS™ Octo Dissociator with Heaters.

The gentleMACS™ Octo Dissociator with Heaters is a benchtop instrument for the fully automated and standardized tissue dissociation or homogenization of up to eight samples. Equipped with eight individual heating units and numerous optimized and ready-to-use gentleMACS Programs the gentleMACS Octo Dissociator with Heaters offers a fully automated workflow for tissue dissociation.

gentleMACS OctoDissociatorThe device is applied for the quantification of cell surface antigens applying flow cytometry. Fresh collected non-human primate tissues are enzymatically dissociated in the gentleMACS™ Octo Dissociator. After isolation of viable tissue cells in a Ficoll gradient centrifugation step, cells are quantified in the flow cytometer, and stained with appropriate antibodies detecting the surface markers of interest. Appropriate in vitro cultured cell lines are included as positive or negative controls.

Accelero has been contracted for this kind of analysis to support a regulatory non-clinical study.
